Nope you may replace player who has not played yet by other player who has not played yet. If you sell player who already played his points will count towards MD1 total score, and yours transfer will be effective in MD2. Example:
You may still sell Lukaku for Ronaldo and Ronaldo's points will be added to your total for MD1. But if you want replace Modrić with KdB, Modrić poinst stay with you for MD1 and KdB points from Belgium game 2 will be added to MD2 total.

You can use 2FT before MD2 but in MD2 can only sub in for 2 unlocked players in MD2. So you can't bring in 2 new players in MD2 for locked players whose team has already played. I think there's not gonna be as much rotation in MD3 as anticipated before tournament starts. WC might be better used after round of 16 after teams are knocked out.

I believe that to be WC wasted. If you play it during md2 you can sell all your locked players with md3 in mind.
